The role of miRNAs in diabetic renal interstitial fibrosis

Synopsis

Diabetic nephropathy is a leading cause of end-stage renal failure, accounting for 35-40% of all new cases requiring dialysis therapy throughout the world. The increasing incidence of diabetes elevates diabetic nephropathy to one of the most important current public health issues, representing a significant burden on the health system. The current existing treatments of diabetic nephropathy can slow its progression but remain suboptimal, so more innovative strategies are needed to prevent and treat this disease. A comprehensive understanding of the mechanisms of nephropathy is essential to develop new strategies. miRNA-based treatment strategies have the therapeutic potential to arrest the progression of diabetic nephropathy and minimise the growing burden of end-stage renal disease. We hypothesise that the in vivo delivery of miRNAs will prevent the interstitial deposition of ECM and the development of renal fibrosis in diabetic nephropathy. This project will determine mechanistically the processes underpinning the actions of miRNAs. Our collective knowledge and experience in both in vitro and in vivo studies will ensure that the role of miRNAs in mitigating diabetic nephropathy will be determined. There is currently a clear gap in effective treatment of diabetic nephropathy that will be addressed by this study.
